Australian Consumers Rate Online Dating Sites | Australian Women ...

According to research commissioned by ratings agency Canstar Blue, one in six Australians have used an online dating site in the last 6 months.

The survey of 2500 people asked Aussies to rate their overall satisfaction with the free and pay for use online dating sites. The research also revealed how the different generations ? Gen Y, Gen X and Baby Boomers ? approach online dating.

Plenty of Fish was the winner of Canstar Blue?s customer satisfaction award, recording the highest five star rating for overall satisfaction as well as for ?ways to communicate?.

Canstar Blue?s Rebecca Logan said ?This is the first year we have looked at Online Dating Sites and our survey showed there were a number of operators in this category ? each of them doing different things well.?

For example, Adult Match Maker received Canstar Blue?s five star rating for the number and relevance of matches and Oasis Active received five star ratings for value for money and site navigation.

The survey also showed Generations Y (35%) and X (34%) were twice as likely as Baby Boomers (17%) to have met their current partners through an online dating site.

?The younger generations also either have, or are more willing to admit to having, what could be described as ?creative? strategies for selling themselves,? said Rebecca Logan.

Canstar Blue found the younger their respondents were, the more likely they were to have used a photo-shopped image (Generation Y ? 24%, Generation X ? 21%, Baby Boomers ? 11%) and exaggerated or lied when writing their online dating profile (Generation Y ? 32%, Generation X ? 23%, Baby Boomers ? 13%).

Generations Y (44%) and X (46%) were also more likely than Baby Boomers (31%) to be looking for casual liaisons over long-term relationships when using online dating sites.

U.S. Department of State Issues Travel Warning for Algeria | Travel ...

Due to continuing terrorist threats, the Department of State has updated its travel warning for all U.S. citizens traveling to Algeria. They recommend that all citizens carefully consider the risks associated with travel to Algeria, including bombings, ambushes, and other terrorist attacks. The threat of kidnappings has also increased, prompting the Department of State to issue a statement of Worldwide Caution [http://www.travel.state.gov/travel/cis_pa_tw/pa/pa_4787.html] on July 18, 2012 to all U.S. citizens traveling abroad.

Most terrorist attacks in Algeria occur in the areas east and south of Algiers, however, extreme caution should be observed for all overland travel throughout the country. U.S. citizens traveling in Algeria should prepare for emergency situations by maintaining reserves of medicine, water, and food in case of an attack. When traveling in cities, citizens are warned to avoid large crowds or demonstrations and to remain aware of their surroundings at all times. Visitors to Algeria are encouraged to stay in hotels with proper security, maintain a low profile, and avoid predictable travel patterns. All U.S. visitors or residents of Algeria can receive travel updates and updated security information by enrolling in the Department of State?s?Smart Traveler Enrollment Program?(STEP). This program also makes it easier for the Department of State to contact them in case of an emergency. Travelers can also stay informed through the Department of State?s Bureau of Consular Affairs website, Facebook or Twitter pages, or free Smart Traveler App, available for the iTunes or Android market. Travelers can find updated information by calling?1-888-407-4747?toll-free in the United States and Canada or, for callers outside the United States and Canada, on a regular toll line at?1-202-501-4444, Monday ? Religion is the Problem

?I also want to say that Islam, beyond the exaggerations, points the finger at something real: under the guise of freedom, in the West we tend to ridicule religion. In the days of his visit in Lebanon, the Pope spoke of violence in words and in deeds. If we want to free the world from violence, we must also free ourselves from the violence of words, from this strong way of offending religion. Unfortunately, the Christians of the West are submissive and unresisting in the face of insults to Christianity.? (Samir Kalil Samir, writing in the AsianNews and reported from Beirut on 9/23/12)

Father Samir, a Catholic priest in Lebanon, has cut to the bone of the most significant problem in international inter-religious dialogue. But reflecting, as he does, the Vatican?s position he fails to understands the modern imperative of freedom of expression.

Religion is often ridiculed in the West, among other reasons, because for so many centuries, preceding indeed the domination of Western religion by Christianity, religion was an abusive and coercive force in society. It was a state dominated means of manipulating symbols to monopolize control of society?s narrative and thereby to control the population. Freedom of conscience was a necessary casualty in this defense of power. Freedom of expression (found in the Roman satirists) was a means of puncturing the pretenses of civil religion and gaining some space for freedom to think independently.

The early Christians, who embraced martyrdom rather than bow to the icons representing Roman imperial power understood this. When those icons were replaced by the cross the Catholic and Orthodox churches, now beneficiaries of this coercive power, quite naturally ceased resisting and embraced it.

Of course the exploitation of civil religion as a tool to achieve or secure political power by an emperor, king, or dictator is quite comprehensible. For the pre-modern state religion, whether it was Christianity or Islam or Buddhism or some Vedic cult, was an important symbol of the unity of the state and society, a source of social mores, an explanation of and justification for the existing hierarchy, and was thus a bulwark of state power. It was natural that the state supported and protected this civil religion, arguably for the good of all those who lived within it and benefited from its religious, cultural, and social cohesion.

The problem in this system, a problem that animated the turn from pre-modernity to modernity, was the way in which this alliance of state power and civil religion constrained personal freedom of conscience. The emerging understanding of humanity found in modernity elevated the free individual?s conscience, and its unconstrained expression, to be the fullest expression of human personhood. This new understanding of humanity was immediately at odds with state supported civil religion, and religious reformation and political revolution necessarily followed.

In a modern understanding, the task of the state is NOT to protect the civil religion, its institutions and its symbols, and with it the cohesiveness of society. The task of the state IS to protect the freedom of individual conscience, and necessarily following that the freedom of individual expression. And this freedom must be protected even if it is expressed as a rhetorical or symbolic ?attack on the civil religion of the state as represented by flags and other iconic symbols. Do Muslims seriously think that the US government will protect the religious symbol of the Qur?an when it freely allows its citizens to burn the U.S. flag?

In other words we are not facing a problem of protecting the religious symbols and avoiding offending those who hold them dear. We are facing a conflict two visions of society:? One in which the state is the defender of a civil religion which insures personal meaning and social order. And another in which the state is regarded as the defender of personal freedom of conscience and expression.? In one vision, it is institutions which have rights and responsibilities.? In another, only individuals; sovereignty, indeed, abides in the people.

This conflict isn?t driven by whether or not Arabs and Muslims easily take offense, or tend toward violent behavior, or some other supposed distinction. It is driven by different understandings of what it means to be fully human and to know one?s self as fully human. There are pre-modern people in every modern state urging that it use the state?s coercive power to protect their identity and social cohesiveness by protecting through civil and criminal law the symbols and powers of a civil religion. And in every state there are those who are modern and will not rest until their own personal freedom of conscience ? and that of their fellow citizens ? is manifest by the state by protecting the fullest freedom of personal expression.

The future of all our societies will depend on how conflicts between these groups are managed, if not resolved, and on the common understanding of the human person that emerges in our increasingly global culture. In the meantime we should not expect that mutual knowledge will lead to mutual understanding and respect, or even acceptance. The differences are profound, the stakes are high, and the good fences that might make good neighbors are falling faster and faster.

Egypt: 14 get death sentences for Sinai attacks

CAIRO (AP) ? An Egyptian court on Monday sentenced 14 members of an extremist group to death by hanging for attacks against police in the Sinai Peninsula, ruling that they were members of an organization that considers even other Islamists to be infidels.

Six of the men were present to be sentenced by the court in the Suez Canal governorate of Ismailia that borders Sinai, while another eight are still fugitives and convicted in absentia.

The death sentences highlight the conflict between the government of President Mohammed Morsi, who hails from the more moderate Muslim Brotherhood, and extremist networks like el-Tawhid wi el-Jihad, blamed for the deadly attack last year in northern Sinai's el-Arish city.

The June 2011 attack against el-Arish's main police station and a nearby bank killed a civilian and a number of police and military officers. The group was also found guilty of storming el-Arish's police station and of smashing statues of former President Anwar Sadat who was assassinated in 1981 after signing Egypt's peace treaty with Israel.

El-Tawhid wi el-Jihad is part of a larger fringe trend in northern Sinai known as Takfir wil-Hijra. The movement's members, dubbed "Takfiris", lead secretive, isolated lives where anything and anyone that does not adhere to their rigid interpretation of Islam is deemed heretical ? including the government and its security forces. Takfir wil-Hijra's leader was executed under Sadat in 1978.

The court issued its final verdict Monday after Egypt's top religious cleric approved the executions, as is customary for death sentences under the nation's legal system.

Another six men involved in the case were sentenced to life in prison. Four others were found not guilty.

Extremist militants operate in disparate groups in the Sinai Peninsula and are believed to have grown in numbers since last year's political upheaval following the ouster of Hosni Mubarak, largely due to a negligible police presence in the area. The volatile region, which links Egypt's borders with the Gaza Strip and Israel, has become increasingly lawless over the past year.

In a brazen attack, unidentified militants killed 16 Egyptian soldiers near the border in August. More recently on Friday, heavily armed militants wearing explosive belts opened fire on Israeli soldiers near the border, killing one.

Rihanna Re-Follows Chris Brown on Twitter (@rihanna @chrisbrown)

September 22, 2012 | 8:30 PM??? Written By: Pierce Watson

Since their surprise kiss at the VMAs, Chris Brown and Rihanna have been on much better terms. Rihanna made it clear they're cool now when she recently re-followed Breezy on Twitter, after un-following him earlier in the year because of a perceived diss track.

Moments after following Brown, Rihanna retweeted a message which seemed suspiciously akin to her situation.

"@EMOT1ONS: I miss you. I miss us. I miss having you to talk to whenever I wanted. #1 love"

It's impossible to guess what will happen next in the Chrianna saga, but as far as we know, they're at least close enough to kiss.

Giant panda cub that died at U.S. zoo had abnormal liver

(Reuters) - The death of a six-day-old giant panda cub at the National Zoo in Washington - an agonizing blow to wildlife conservation efforts - could be linked to an abnormal liver, the zoo's chief veterinarian said on Monday.

A day after the zoo's staff failed to revive the cub with lifesaving measures, including CPR, the initial results of an examination showed the panda was likely a female whose body displayed no obvious signs of trauma or infection.

"Her coat was beautiful. It was in very good condition," veterinarian Suzan Murray said at a press conference.

The cub was born on September 16 but had not yet been given a name, in line with a Chinese tradition that pandas are not named for 100 days. The arrival was cause for celebration among zoo officials and wildlife conservationists given the daunting odds for the endangered species reproducing in captivity.

Fewer than 1,600 giant pandas are known to exist in the wild, and about 300 live in zoos and wildlife centers around the world. Breeding is a critical challenge. One in five cubs born in captivity die in their first year of life, the National Zoo said on its website.

Preliminary results of an examination of the cub show the tiny animal had a discolored liver, which was hardened in some areas, Murray said.

"This can suggest there was a liver component to the death," Murray said.

Another unusual finding, according to Murray, was fluid in the cub's abdomen, normal for adult pandas but odd for a juvenile. Pathologists were analyzing the fluids, Murray said.

"The amount appears moderate to increased for such a small cub," she said, adding that further testing was necessary to confirm a cause of death.

The body of the cub weighed about 3.5 ounces (100 grams).

Zoo officials said they were concerned about the reaction of the cub's mother, Mei Xiang, who had given birth after years of failed efforts at conception, including five "pseudo pregnancies" in which her hormone levels and behavior indicated she was carrying a cub when she was not.

Her first cub, Tai Shan, was born July 9, 2005. Tai Shan is now at the China Conservation and Research Center for the Giant Panda in Wolong.

The zoo has used a Panda Cam to monitor Mei Xiang, who appeared to sleep well Sunday night and who ate her regular diet of bamboo, fruit and biscuits, the zoo said on its website.

"Watchers noticed her cradling an object, as she did before the birth of the cub. Scientists and keepers believe this is an expression of her natural mothering instinct," according to the zoo's website.

The cub's death was discovered Sunday morning after panda keepers and zoo volunteers heard a distress sound from Mei Xiang.

"We want to make sure that her behavior adjusts back to normal," said zoo spokeswoman Emily Grebenstein.

Dr. Fischer's Game Night: War of the Rings Card Game- Still Fun

In the aftermath of our summer vacation I was left thinking about which games I've purchased that have actually gotten a lot of play. I'm as guilty as most gamers of accumulating vast piles of lead, rules, and boardgames that then sit sad and forlorn on the shelf waiting to be played or painted. This year the kids are off to school, however, so we are on a tight budget and I am trying to get the dusty stuff off the shelves and onto the table.

I picked up the Lord of the Rings Card Game a while back with high hopes. It's cooperative, the components are beautiful, and the theme is popular. When I played it at the game store it was a lot of fun. Unfortunately the rules seem to have been written in one language and then translated across two more before making their way into English. As a result our first attempts to play at home were just demoralizing. I put it away and only took it out again for the vacation.

When the kids text "RTFM" to each other they are being wise indeed. When I took the time to really read the instructions and understand them much of the game fell into focus. We ended up playing several times and had a lot of fun. Much of what I said in my original review still holds true now. I think what strikes me in hindsight is how fun it is to come close to winning and then get crushed. It sounds silly but a challenging coop game where you Almost win is really a good time. The game drags you back for another go.

Further, the factions you can play in the game allow each player to find something they enjoy doing. My wife enjoys dealing out damage and slaying monsters. There's a faction for that. I like cheating fate and disrupting the plans of evil. There's a faction for that as well. The game allows and supports varied play style.

Finally, the game has a nice narrative quality. As you struggle to complete the quests you do feel like a story is being told. There are narrow escapes, epic battles, and through it all you wait for Gandalf to appear and pull your bottoms out of the fire! Which will happen, to everyone's relief.

The Lord of the Rings Card Game has absolutely held up over time. I recommend reading the rules a few times and struggling through a game or two. At that point I suspect you'll be hooked.

DIY Project Parade ? a place to share and search for DIY creativity ...

I love this time of the week ? a chance to see ?your creativity, your crafts, your DIY projects, your home improvement. I love visiting the links and sharing our common interest: DIY!

Last week, along with a few other things, I shared the bathroom makeover progress from the apartment renovation:

how to tile a bathroom floor

Some good things about this past week?

DIY:

 • Apartment floors are finished! Ohhh and they?re sooo pretty! Just you wait and see! I?m definitely doing a happy dance over this accomplishment. Ugh. Lots of work! So glad it?s over!
 • Blinds are hung?freebies too. Mr. DIY dug them out from somewhere and I?m sure he told me where, but I think I was probably working and only heard the important part ? free blinds. And ?he? installed them. Thanks, babe!
 • New deadbolt installed. My impatience had me putting it right back in the box and thinking the brassy gold deadbolt with lost key and the fact that it didn?t match the brushed nickel door knob really wasn?t that important. Again, Mr. DIY rescued the day. He?s my hero.
 • Dressing room area of the apartment bedroom started. So fun!
 • Few paint touch ups completed. Since fall has arrived and the window air conditioner was no longer needed {brrrrr!}, it was removed and I could paint the window sill. And a ginormous box on the apartment wall {something for an outdated telephone line} was removed and I touched up that area too.
 • Old cheap bar stools and a little kitchen island/bar were given a little DIY magic. Such a budget friendly way to furnish the apartment since we already had this stuff just sitting around collecting dust. Saved some $!
 • Dug out some fall decorations and I?m starting to play with getting festive for fall.

Not DIY related:

 • Met up with a sweet blog friend. {Reposhture}. It?s so good talking and laughing with a friend who shares the same interests.
 • Fun fashion photo shoot where I pretended to be a photographer. I admit?I loved the little break away from months of non stop DIY.
 • Invitation to participate in the local Symphony Holiday Tour of Homes fundraiser! Such an honor!

But, life isn?t perfect and there were some pretty crappy things this week too. At times like those, I hold a pity party for one for a bit. Then I count my blessings.?

What I love about today is it signals a brand new week and a clean slate for me. I love this day when I can cancel the pity party and celebrate what?s been going on in your life as I invite you to share my favorite topic. DIY! Thanks so much for being here today, friends!

Show off your latest DIY:?

2. This blog linky party is for?DIY projects only. Links not related to DIY will be deleted. Please share your link one week only.??Please do not link recipes, giveaways, linky parties, promotions or a collection of inspiration.?

Awesome projects with great pictures may be included in a post of DIY Project Parade highlights later in the week, featured on DIY Show Off, Facebook,Twitter, G+ or Pinterest. Look for the DIY Project Parade highlights this week!

If you have a fall related project, don?t forget to come back Wed. to link up to the DIY Fall Festival and again on Friday when I share my fall mantel and host the Countdown to Fall linky party. Lots of partying this week, friends!

iPhone 5 Fans Want More Features, Still Line Up [Video]

NEW YORK CITY ? If you?re an Apple fan, nothing is going to dissuade you from getting an iPhone 5. That?s what TechNewsDaily discovered on the streets of Manhattan ? and in Grand Central Station ? this morning (Sept. 21) shortly before the Apple stores opened at 8 a.m.

Diehard campers notwithstanding, most of the people we met had been waiting for just a few hours, typically from about 5 a.m. or a bit earlier. At the iconic glass-cube Apple store on Fifth Ave., some fans said they had lined up simply for the excitement of being there. Fans stuffed into a hallway at Grand Central were more subdued.?

There was no question of getting anything but an iPhone. Our offers of a free Samsung Galaxy S3 were scoffed at.

Nevertheless, many people saw room for improvement. One fan was disappointed that the?phone?didn?t have NFC chips, which allow users to tap their phones to a payment terminal, taking the place of credit cards or transit passes. Another customer said he?d like the ability to tap phones to exchange photos wirelessly. The Samsung Galaxy S3 offers both features.

Top U.S. cancer hospital plans 'moonshot' against disease | The Salt ...

Top U.S. cancer hospital plans ?moonshot? against disease

MD Anderson ? Houston hospital to spend$3 billion over 10 years to jump-start research.

The nation?s largest cancer center is launching a massive "moonshot" effort against eight specific forms of the disease, similar to the all-out push for space exploration 50 years ago.

The University of Texas MD Anderson Cancer Center in Houston expects to spend as much as $3 billion on the project over the next 10 years and already has "tens of millions" of dollars in gifts to jump start it now, said its president, Ronald DePinho.

One of the cancers is myelodysplastic syndrome. "Good Morning America" co-anchor Robin Roberts has that kind and had a bone marrow transplant to treat it on Thursday. The others are especially deadly forms of breast and ovarian cancer, along with lung, prostate, melanoma and two types of leukemia.

The project aims to find cures and lower deaths. Although no overall benchmarks have been set, individual projects for various cancers have specific goals.

With genetic information and more precise drugs, "we have many of the tools we need to pick the fight of the 21st century" and find ways to defeat these cancers, DePinho said.

Otis Brawley, chief medical officer of the American Cancer Society, which has no role in the project, praised DePinho?s effort.

"I?m thrilled to see somebody take the lead," Brawley said. "The results that I see him promising, in my mind are reasonable," both in terms of raising money and fighting cancer.

Cancer death rates have been falling since the 1990s at an average of more than 1 percent per year but the disease remains a top killer worldwide. In the United States this year, estimates are that more than 1.5 million people will be diagnosed with cancer, and more than 500,000 will die from it.

The MD Anderson program was inspired by the goal President John F. Kennedy announced in 1962 to put a man on the moon by the end of that decade. He described it to Congress that May and in a speech in September at Rice University, a mile from MD Anderson.

The Houston cancer center treats 112,000 patients a year and has been building a database of tumor samples and their genetic characteristics, especially of breast and ovarian cancers.

story continues below

A year ago, when DePinho became its new president, he started a competition among its researchers to submit ideas for how to make fresh inroads against the disease. Six teams were chosen to tackle the eight cancers.

Each team has specific goals, ranging from basic research to clinical trials that test treatments, biomarkers and diagnostics. Some projects are aimed at novel prevention methods ? a mandatory role for any of the federally funded National Comprehensive Cancer Centers, DePinho said.

The teams will focus on personalizing treatment according to an individual?s tumor genes, real-time assessment of the effectiveness of therapies being tried, better diagnoses and early detection and reducing side effects of treatment.

For example, one project will jointly target a deadly type of ovarian cancer and "triple-negative breast cancer" ? breast tumors whose growth is not fueled by estrogen, progesterone or the gene that the drug Herceptin targets. The cancers share some striking similarities at the genetic and molecular level and treatments for them are converging, MD Anderson scientists say.

Scientists plan to study one cancer pathway and how nearby tissue influences the cancer?s growth. They also hope within five years to find a way to detect ovarian cancer early. Experts have recommended against routinely screening health women for ovarian cancer now because methods to do so are too flawed.

Money for all of the moonshot projects will come from foundations, gifts from individuals, grants, revenues from treating the additional patients the center expects to attract, and patents and royalties from discoveries, MD Anderson officials said. That will be on top of the $700 million the cancer center spends each year now on research.

How the sub-Saharan cheetah got its stripes: Californian feral cats help unlock biological secret

ScienceDaily (Sep. 20, 2012) ? Feral cats in Northern California have enabled researchers to unlock the biological secret behind a rare, striped cheetah found only in sub-Saharan Africa, according to researchers at the Stanford University School of Medicine, the National Cancer Institute and HudsonAlpha Institute for Biotechnology in Huntsville, Alabama. The study is the first to identify a molecular basis of coat patterning in mammals.

The scientists found that the two felines share a biological mechanism responsible for both the elegant stripes on the tabby cat and the cheetah's normally dappled coat. Dramatic changes to the normal patterns occur when this pathway is disrupted: The resulting house cat has swirled patches of color rather than orderly stripes, and the normally spotted cheetah sports thick, dark lines down its back.

"Mutation of a single gene causes stripes to become blotches, and spots to become stripes," said Greg Barsh, MD, PhD, emeritus professor of genetics and of pediatrics at Stanford and an investigator at the HudsonAlpha Institute.

The differences are so pronounced that biologists at first thought that cheetahs with the mutated gene belonged to an entirely different species. The rare animals became known as "king cheetahs," while affected tabby cats received the less-regal moniker of "blotched." (The more familiar, striped cat is known as a mackerel tabby.)

The study will be published Sept. 21 in Science. Christopher Kaelin, PhD, a senior scientist in the Barsh laboratory, is the co-first author; Xiao Xu PhD, from the National Cancer Institute-Frederick National Laboratory for Cancer Research and the Sichuan Key Laboratory of Conservation Biology on Endangered Wildlife in Sichuan, China, is the other co-first author. Marilyn Menotti-Raymond, PhD, of NCI-Frederick is the senior author.

Barsh and his lab members have spent decades investigating how traditional laboratory animals such as mice develop specific coat colors. His previous work identified a variety of biologically important pathways that control more than just hair or skin color, and have been linked to brain degeneration, anemia and bone marrow failure. But laboratory mice don't display the pattern variation seen in many mammals.

"We were motivated by a basic question," said Barsh of the turn to the study of big (and little) cats. "How do periodic patterns like stripes and spots in mammals arise? What generates them? How are they maintained? What is their biological and evolutionary significance? It's kind of surprising how little is known. Until now, there's been no obvious biological explanation for cheetah spots or the stripes on tigers, zebras or even the ordinary house cat."

The research relied primarily on DNA samples from feral cats in Northern California captured for sterilization and release, on tissue samples provided by the City of Huntsville Animal Services group, and on small skin biopsies and blood samples from captive and wild South African and Namibian cheetahs. It also hinged on the recent availability of the whole-genome sequence of the domestic cat. (Menotti-Raymond's research focuses on the genomic analysis of the domestic cat to better understand many human diseases.)

"The Laboratory of Genomic Diversity at the National Cancer Institute has long championed the cat as an animal model of human disease," said Menotti-Raymond. "Studying color variation in cats provides the opportunity to uncover new principles of gene action and interaction that may have unexpected applications to understanding developmental and morphologic variation in natural populations, including humans."

Comparing gene sequences of feral cats with different patterns allowed Kaelin and Xu to identify mutations in a gene they dubbed Taqpep associated with the blotched tabby markings: 58 of 58 blotched tabbies had a mutation in each of its two copies of Taqpep, while 51 of 51 mackerel tabbies had a least one unmutated version.

Taqpep encodes a protease normally found in the cell membrane, but that can also be cleaved to allow it to diffuse outside the cell. This ability to float freely and interact with other molecules in the extracellular soup is a key component of a principle called reaction diffusion proposed by the famous computer scientist Alan Turing, PhD, in 1952 as a way to explain how periodic patterns (like stripes and spots) can arise out of randomness.

"Turing realized that, under specific conditions, diffusible 'activator' and 'inhibitor' molecules can self-organize into a variety of periodic patterns," said Barsh. "We are excited about the idea that Taqpep might be an entry point to understand if, and how, reaction-diffusion mechanisms can explain 'how the leopard got its spots.'"

After nailing down Taqpep's role in tabby stripes (and analyzing its sequence in more than 350 other cats of 24 distinct breeds), Kaelin wondered if it might play a similar role in generating and maintaining the spots on wild and captive cheetahs. He obtained blood samples from a king cheetah named Kgosi, a resident of a wildcat education and conservation program in Northern California, and found that Kgosi also had a mutation in Taqpep.

Kaelin next contacted Ann van Dyk, who maintains a cheetah conservation center in South Africa from which all captive king cheetahs, including Kgosi, originate. (Van Dyk was the first to learn, though meticulous breeding records, that the king cheetah pattern is due to a recessive genetic mutation.) Van Dyk obtained DNA samples from all her cheetahs, allowing confirmation that a Taqpep mutation is responsible for the king cheetah pattern.

Mammals aren't the only animals with patterned hair or skin, obviously. Fish, salamanders and some invertebrates also have stripes and spots. However, there is an essential difference. While the non-mammals simply add stripes or spots as they grow to adulthood, mammals keep the same number and pattern by increasing the surface area of the contrasting colors.

"Somehow, cells in the black stripes know they are in a black stripe and remember that fact throughout the organism's life," said Barsh. "We were curious about what's happening at the boundary between light and dark stripes and spots. How do these spots know to grow with an animal?"

When Kelly McGowan, MD, PhD, a senior scientist in Barsh's group, studied fetal cat skin after seven weeks of gestation, she found that the tabby pattern begins to arise only when the hair begins to grow. In other words, there are no apparent differences between the cells themselves -- only in the color of hair they produce. That suggested that the changes in color are due to differences in the levels of expression of certain genes within the cells.

Lewis Hong, a former graduate student in Barsh's lab, used a technique he developed called EDGE to identify changes in gene expression levels between black and yellow areas of cheetah skin (obtained under anesthesia). He found several differences, many associated with a pathway influencing the expression of a gene called Edn3. McGowan found that Edn3 mRNA was produced at the base of the follicles making the black hairs. To test their theory, the researchers collaborated with a group at Florida International University to study a yellow-colored laboratory mouse that had been engineered to express Edn3. The coats of the resulting animals were much darker than their unmodified peers.

"This is very strong evidence that Edn3 is a critical regulator of black versus yellow hair in animals," said Barsh. The researchers hypothesize that expression of Taqpep is required to establish a pattern of stripes or spots in early feline development that is then carried out by Edn3 as the hair grows.

Clearly, not all cats are patterned. In particular, some big adult cats like African lions and mountain lions are distinctive for their lack of color variation even though their cubs are striped. Furthermore, Taqpep mutations are surprisingly common in some non-striped domestic cat breeds like the Abyssinian and the Himalayan.

"We know there's a mutation that suppresses pattern formation in some cats," said Barsh. "We'd like to investigate that mechanism as well."

In addition to Kaelin, Barsh, McGowan and Hong, Stanford researcher and technician Hermogenes Manuel also participated in the study.

The research was funded by the National Institutes of Health, the NCI and the HudsonAlpha Institute.
